    Validation of the Oregon Scientific BPU 330 for self-monitoring of blood pressure according to the International Protocol

The Oregon Scientific BPU 330 blood pressure monitor is an electronic device for upper arm measurement. This study assessed the accuracy of the Oregon Scientific BPU 330 blood pressure monitor according to the International Protocol by the Working Group on Blood Pressure Monitoring of the European Society of Hypertension for validation of blood pressure measuring devices.Method: 52 participants over 30 years of age were studied in the validation. Nine blood pressure measurements were taken alternately with a mercury sphygmomanometer by two observers, and by the supervisor, using the BPU 330 device. A total of 33 participants were selected for the analysis. The validation was divided into two phases. Phase 1 included 15 participants. If the device passed phase 1, 18 more participants were included. The 99 pairs of measurements were compared according to the International Protocol. The device was given a pass/fail recommendation based on its accuracy compared with the mercury standard (within 5, 10, and 15 mmHg), as well as the number met in the ranges specified by the International Protocol.Results: The mean and standard deviation of the difference between the mean of the observers and the BPU 330 device were 1.7 ± 4.7 mmHg and 2.8 ± 3.9 mmHg for systolic blood pressure (SBP) and diastolic blood pressure (DBP), respectively. In phase 1, the device passed with a total of 33, 43, and 44 SBP readings; 38, 44, and 45 DBP readings were within 5, 10, and 15 mmHg, respectively. In phase 2.1, 81, 95, and 96 for SBP, and 83, 95, and 98 for DBP readings fell within the zones of 5, 10, and 15 mmHg, respectively. In phase 2.2, the last phase, 28 participants fell within the zone of two of the three comparisons, lying within 5 mmHg for SBP and 29 participants for DBP. No participants fell within the zone of all three of their comparisons over 5 mmHg apart for both SBP and DBP.Conclusion: The BPU 330 can be recommended for self-monitoring of blood pressure in the adult population, according to the International Protocol.Keywords: blood pressure, self-monitoring, hypertension, International Protoco

    Evaluation of Ecological Construction in Kunming using the DPSIR Model

    Kunming is located in the south-western part of China and a lakeside city situated on plateau. Its ecological construction is typically representative. This paper takes the ecological construction data of Kunming City from 2007 to 2016 as a sample, and uses the Driving Force-Pressure-State-Influence-Response (DPSIR) model to quantitatively analyze the date, aiming at an objective evaluation of the future ecological construction of Kunming. The urban road greening proposes practical measures. The results of research show that in the past ten years, Kunming's ecological construction has shown a ā€œfast-slow-stableā€ upward trend, indicating that it has achieved remarkable results. In the future, importance will continue to be attached to the development of tertiary industry, focusing on the introduction of innovative talents, and encouraging the active participation of the citizens. This is the ā€œthree-horse carriageā€, which means three horses pull the cart together, for the sustainable development of ecological construction in Kunming

    Construction of CaF2-appended PVA nanofibre scaffold

    In this work, a new material, calcium fluoride ( CaF2 )-appended poly(vinyl alcohol) (PVA) nanofibre scaffold, was prepared through electrospinning technique successfully. Scanning electron microscopy result showed that the morphology of the fibres was uniform and smooth, and the average diameter of the fibres was about 200 nm. Transmission electron microscopy results showed that many CaF2 nanoparticles were well dispersed in the PVA fibre matrix. The water-resistant ability of the scaffold was improved through intermolecular crosslinking of PVA by formaldehyde vapour. This novel material seems to be a promising scaffold for bone tissue engineering

    The Use of Geographically Weighted Regression for the Relationship among Extreme Climate Indices in China

    The changing frequency of extreme climate events generally has profound impacts on our living environment and decision-makers. Based on the daily temperature and precipitation data collected from 753 stations in China during 1961ā€“2005, the geographically weighted regression (GWR) model is used to investigate the relationship between the index of frequency of extreme precipitation (FEP) and other climate extreme indices including frequency of warm days (FWD), frequency of warm nights (FWN), frequency of cold days (FCD), and frequency of cold nights (FCN). Assisted by some statistical tests, it is found that the regression relationship has significant spatial nonstationarity and the influence of each explanatory variable (namely, FWD, FWN, FCD, and FCN) on FEP also exhibits significant spatial inconsistency. Furthermore, some meaningful regional characteristics for the relationship between the studied extreme climate indices are obtained

    Genotypic and Phenotypic Characterization of IncX3 Plasmid Carrying blaNDM-7 in Escherichia coli Sequence Type 167 Isolated From a Patient With Urinary Tract Infection

    Infections due to New Delhi metallo-beta lactamase (NDM)-7-producing Escherichia coli are infrequent and sporadic. In this study, we report one case of recurrent urinary tract infection caused by blaNDM-7-producing E. coli belonging to phylogenetic group A, sequence type (ST) 167. In this study, we aimed to describe the genotype and phenotype of blaNDM-7-producing E. coli in China. The isolate exhibited resistance to Ī²-lactam antimicrobials, trimethoprim-sulfamethoxazole, quinolones, and aminoglycosides. blaNDM-7 is located on a conjugative plasmid designated pJN05NDM-7 belonging to type IncX3. pJN05NDM-7 was fully sequenced and compared with all publicly available blaNDM-7-harboring plasmids. pJN05NDM-7 is almost identical to pKpN01-NDM7 and pKW53T, although the plasmids are geographically unrelated. The comparison of IncX3 plasmids harboring blaNDM in China showed high similarity, with genetic differences within insertion fragments. Notably, the differences in plasmids of animal and human origin were insignificant, because only one plasmid showed deletion inside the ISAba125 region compared with pJN05NDM7. Our study demonstrates that E. coli carrying IncX3 plasmids play an important role as a reservoir and in the spread of blaNDM. Further studies should be performed to control the dissemination of blaNDM among food animals

    Study on the vibration characteristics of structural of hydrocyclone based on fluid structure interaction

    The hydrocyclone of main diameter 28mm with a typical variable diameter pipe structure was selected as the research object, and the interaction of the fluid and the structure was considered on the basis of the hypothesis of small deformation. The two-way fluid structure interaction (FSI) model of the hydrocyclone with variable diameter pipes was presented in this paper, and the related numerical simulation and experimental research was also carried out. The structure vibration characteristics of the structure were analyzed base on the fluid structure interaction, and the vibration characteristics of the structure were obtained. The vibration characteristics of the between the results of numerical simulation and experimental were consistent. Through above analysis showed that the two-way FSI calculation model of hydrocyclone with variable diameter pipe was reasonable. The influence of coupling effect on flow field couldnā€™t be ignored. The interaction between fluid and structure changed the movement characteristics of the structure, which should be considered in the design and application of hydrocyclone

    Autophagy Inhibitor LRPPRC Suppresses Mitophagy through Interaction with Mitophagy Initiator Parkin

    Autophagy plays an important role in tumorigenesis. Mitochondrion-associated protein LRPPRC interacts with MAP1S that interacts with LC3 and bridges autophagy components with microtubules and mitochondria to affect autophagy flux. Dysfunction of LRPPRC and MAP1S is associated with poor survival of ovarian cancer patients. Furthermore, elevated levels of LRPPRC predict shorter overall survival in patients with prostate adenocarcinomas or gastric cancer. To understand the role of LRPPRC in tumor development, previously we reported that LRPPRC forms a ternary complex with Beclin 1 and Bcl-2 to inhibit autophagy. Here we further show that LRPPRC maintains the stability of Parkin that mono-ubiquitinates Bcl-2 to increase Bcl-2 stability to inhibit autophagy. Under mitophagy stress, Parkin translocates to mitochondria to cause rupture of outer mitochondrial membrane and bind with exposed LRPPRC. Consequently, LRPPRC and Parkin help mitochondria being engulfed in autophagosomes to be degraded. In cells under long-term mitophagy stress, both LRPPRC and Parkin become depleted coincident with disappearance of mitochondria and final autophagy inactivation due to depletion of ATG5-ATG12 conjugates. LRPPRC functions as a checkpoint protein that prevents mitochondria from autophagy degradation and impact tumorigenesis

    The role of p53 in the DNA damage-related ubiquitylation of S2P RNAPII

    DNA double-strand breaks are one of the most deleterious lesions for the cells, therefore understanding the macromolecular interactions of the DNA repair-related mechanisms is essential. DNA damage triggers transcription silencing at the damage site, leading to the removal of the elongating RNA polymerase II (S2P RNAPII) from this locus, which provides accessibility for the repair factors to the lesion. We previously demonstrated that following transcription block, p53 plays a pivotal role in transcription elongation by interacting with S2P RNAPII. In the current study, we reveal that p53 is involved in the fine-tune regulation of S2P RNAPII ubiquitylation. Furthermore, we emphasize the potential role of p53 in delaying the premature ubiquitylation and the subsequent chromatin removal of S2P RNAPII as a response to transcription block

    Targeting Radioresistant Breast Cancer Cells by Single Agent CHK1 Inhibitor via Enhancing Replication Stress

    Radiotherapy (RT) remains a standard therapeutic modality for breast cancer patients. However, intrinsic or acquired resistance limits the efficacy of RT. Here, we demonstrate that CHK1 inhibitor AZD7762 alone significantly inhibited the growth of radioresistant breast cancer cells (RBCC). Given the critical role of ATR/CHK1 signaling in suppressing oncogene-induced replication stress (RS), we hypothesize that CHK1 inhibition leads to the specific killing for RBCC due to its abrogation in the suppression of RS induced by oncogenes. In agreement, the expression of oncogenes c-Myc/CDC25A/c-Src/H-ras/E2F1 and DNA damage response (DDR) proteins ATR/CHK1/BRCA1/CtIP were elevated in RBCC. AZD7762 exposure led to significantly higher levels of RS in RBCC, compared to the parental cells. The mechanisms by which CHK1 inhibition led to specific increase of RS in RBCC were related to the interruptions in the replication fork dynamics and the homologous recombination (HR). In summary, RBCC activate oncogenic pathways and thus depend upon mechanisms controlled by CHK1 signaling to maintain RS under control for survival. Our study provided the first example where upregulating RS by CHK1 inhibitor contributes to the specific killing of RBCC, and highlight the importance of the CHK1 as a potential target for treatment of radioresistant cancer cells
